for measuring of the activity of enzyme

I don't know why should we must do the procedure of protein precipitation using HPO3 before we measure the activity of Enzyme.I am measure the activity of SDH,but I can not find any activity of SDH in the lymphocytes, why? Thank you very much, I am looking for your kind help.

First things first, you don't need to post the same thing 3 times, put it all together and post it once.
Also some more information would be good.
SDH is really only present in thymic T-cells, ie you need to get a mouse hit it hard over the head and rip out it's thymus (on front of the heart). T-cells in the periphery won't have too much.
You will need to check all this out as I'm a bit rusty.
SDH is also an activation marker, so to get a good result you might consider activating T-cells with a mitogen or perhaps activation beads..

This is all guesswork so I'd recomend some reading in the library.
