how to create cell cultures?

dr. nick - 6-8-2005 at 00:41

Hello everybody,

can anyone provide swim with information on the how-to for creating primary cell cultures, especially from/for plant cells?

forgive my ignorance, not even sure if this is the right forum, but it fits better than beginnings swim thought.
Not that one couldn't find information on this, far from it, it's much too much, can't filter it ... though most of it isn't for plant cells, and working with trypsin ... argh, very confusing for the beginner.

Thanks for every hint or pointing in the right direction!

Look in the Kirk-Othmer under "Cell Culture Technology"

If you donĀ“t have this then get the torrent here

Hey Organikum,

What do you mean by cell culture?

If you mean tissue culture & you want to do it in home .... here is the 2nd hit in gsr

www.une.edu.au/~agronomy/AgSSrHortTCinfo.html

That procedure seems pretty easy ... i am going to do some experiments using it some day. and it is designed for plants.

Hello
Here is a very good book i got from polverone ... actually .. i've printed it!

Plants from Test Tubes - Intro to Micropropagation - Lydiane Kyte

h**p://www.geocities.com/alnokta1/ebooks/plantsftt.zip

Although it is not the latest it still a fine read

alnokta,

My downloads from the site you mention turn out to be corrupt files. Always. Is the book in the forum library? How did you get it? Could you attach the zip file or upload it somewhere else?

Thanks.

I've forgot to mention that you will need 7-zip to open any file from my site .. i had to rename the files from .7z extension to .zip cause yahoo refuses to upload .7z files ....
They are not corrupt ... just install 7-zip and it will extract just fine....

Thanks solo. Thanks alnokta.

Does anybody has first hand experience with micropropagation?

You will definately need to build a laminar flow hood at some point.
www.plantsciences.ucdavis.edu/eh/Ehweb/dwb/lamflohd.pdf

You want to have a stainless steel bench top for ease of cleaning. Clean it every day it is used with 70% Ethanol before and after use. Use the UV light with the hood off for a minimum of 15 minutes before you plan to use the hood. Do not look at the UV light, or use UV lighting while you are working.
